  • Alpha mangostin isolated from mangosteen rind extract (Garcinia mangostana, L) has good anti-cancer activity but poor solubility and low selectivity against cancer cells are the drawbacks of this isolate. Increasing the effectiveness of alpha mangostin compounds has been made in the form of chitosan alginate polymeric nanoparticles called NANO-AMKAL. Based on the results of In Vitro research showing that NANO-AMKAL is very active as an anticancer breast, it is necessary to carry out an In Vivo test to examine the effectiveness of NANO-AMKAL preparations on breast cancer of Wistar rats (Rattus norvegicus) and obtain an effective dose. This research method uses the injection of 7,12- dimethylbenz(a)anthracene (DMBA) as a carcinogenic agent (20 mg) subcutaneously into the breasts of rats which were divided into 6 treatment groups, namely positive control, negative control, pure alpha mangostin, NANO-AMKAL 5mg, 10mg, and 20mg orally, during the treatment were evaluated for weight and tumor volume every 3 days then surgery was performed on day 14, breast and lung cancer tissue was taken for histopathological studies. NANO-AMKAL succeeded in increasing the activity of alpha mangostin for the treatment of breast cancer in Wistar rats (Rattus norvegicus). NANO-AMKAL with 0.33mg alpha mangostin had a healing effect three times better than 20mg pure alpha mangostin and there was no significant difference with Tamoxifen. The effective dose of NANOAMKAL as anti-breast cancer in Wistar rats (Rattus norvegicus) was NANOAMKAL 20mg, which showed a good healing response and the tumor volume continued to decrease up to 17.43% on the 14th day. This is directly proportional to histopathological tests which showed tissue repair and no metastases.
